Italy in September - need a bit of help

So here's the deal I've been to Rome, Florence, and Venice for a cruise stop back in 2008. My wife has never been to Italy. I know she would like to see all the tourist sites though. We really enjoy relaxing, exploring towns, the beach is also a favorite of ours. For example when we went to Greece last year - we REALLY enjoyed just renting an ATV and beach hoping in Mykonos and Santorini. We were thinking of going to Rome, Venice, Florence (she wants to do a wine tour and leaning tower of Pisa),Amalfi Coast, Milan, and Lake Como. I don't think we can accomplish this in two weeks. If I had to pick and choose which place to go - what do you recommend and why? I plan on going in September for 2 weeks. Do you recommend one city over another or maybe one place during another time of year? Please help! I think the two options are: OPTION 1 doing Rome for 2 days (all day condensed tour of Colosseum, Trevi Fountain, and 3 churches - they have way to many so just limit it.), Florence for 2 (one day for leaning tower of Pisa and one for wine country), Venice for 3, Naples for 1, Amalfi for 5 OPTION 2 doing Rome for 2 days, Florence for 2, Venice for 3, Milan for 2, and Lake como for 4 days.
