Do Shortcuts prevent character unlocks?

So I've been using the 3-1 shortcut to chain-run through temple to try and unlock Valerie, but there doesn't seem to be a coffin spawning anywhere. I couldn't find any mention of it through the wiki or anything, but does using shortcuts prevent the character unlock coffins from spawning in Spelunky 2 or something?
