Question About The Worm Entrance

Okay so I know the entrance comes up in 2-1 and 3-1. But my question is this, if I throw the damsel into The Worm entrance in 2-1 but I don't go with it will the The Worm entrance still spawn in 3-1 for me to take? I guess this leads to another question now can I go to The Worm in 2-1 and 3-1 or only enter it once?
