What size is considered true living soil?

I was curious about flower beds. I am interested in a 2x4 bed. I've heard people say that size isn't enough to truly be considered living soil. I usually just do 5 gallon containers of soil, and most say that is also not enough to he considered living soil. Thoughts on minimum size of flower bed? Is a 2x4 good enough?
