Hey everyone,
I'm new to protein characterization and have a question that's probably quite basic but nonetheless I haven't found clear answers looking in papers/google
I want to express a catalytically inactive protein mutant to assess downstream targets. First I thought I could just overexpress the mutant using a viral vector, but then I will still have the wild-type endogenous protein with normal activity. Do people in general knockout the endogenous protein before expressing mutant proteins? What's the norm?
