Anyone have any luck with this? Obviously A/B would work but losses offset higher IN bioavaliability. Is there a better way?

Extraction / Purification of Ketamine from Compounded Ketamine Meds

Ketamine is a touchy molecule. It likes to be pure. It's a whole different level of therapeutic the more pure it is and the higher bioavailability the route of admin.

This method is relatively easy and yields a purity that is sufficiently wonky, very much like any good quality sugar ketamine.

Please feel free to improve and correct my words and statements here... I'm just a dude with too much time on his hands, for the most part.

Updates at bottom if I every updat.

Materials

KETAMINE Rx - assumed k is in solid preparation, compounded into troche or tablets for oral/sublingual use

at least 20 tablets, depending on dosage. ~1 gram of desired molecules between your tabs as minimum. ex: 100mg troches, use at least 10

100-200ml of 99% Dichloromethane (DCM)

Acetone, dry** and cold (32 or less dg f)

**dry meaning you purchase the acetone anhydrous/acs grade or dry impure acetone yourself with anhydrous Epsom Salt (well baked MGS04)****

**** To dry acetone with mgs04 add 1 gram of anhydrous mgs04 per liter of impure acetone., say hardware store brand (anhydrous mgs04 - purchase from lab supply store OR bake the salt at 500 degrees for 30 mins to make it anhydrous, mortar and pestle the dry salt to powder).

500ml beaker with cover

Distilled water

Drying tray

soft bristle brush (acetone resistant, or a throwaway tooth brush)

Micropipette, Sep funnel or other means to effectively move solvents

optional: your own safety - use gloves and goggles and don't huff acetone, Corky

optional: magnetic stir plate

What's the plan to extract our molecules?

Ketamine is soluble in DCM and water, much more so in water. Sugars, starches, and apparently the entire contents of my compounded products dissolve in the DCM. That's the shit compounds binding and flavoring and denaturing -- and we don't fucking want em - we're trying to abuse ketamine, for fucks sake!

DCM is also not something that mixes with water (think oil floating on water). That water immiscibility combined with varying degrees of molecule solubility in a cocktail of substances between the mingling liquids are what are exploited to isolate and concentrate the ketamine in the solvent it loves more. With the right solvents, the undesirable stuff remains mostly in the junk solvent and the desired molecules slip into and concentrate in the co solvent. DCM is our garbage man and and water is our adorable little ketamine molecule nanny.

  1. Dissolve your compounded trouches, tablets, or whatever you have at about 25-30 units per 100ml DCM... don't crush or chop, let the DCM dissolve in full - it will dissolve them rather quickly. Note that the trouches are best for this for whatever reason (be aware that pharmacy recipes and ingredients vary, prob not by much, but I do not know the extent).
  2. add 100ml of distilled water to beaker with DCM and dissolved tablets
  3. Swirl or magnetically stir the water and DCM for about 10 mins at vigorous pace, but don't get overzealous - DCM tends to produce emulsions and "bubbles" of unclean separation can make isolating the ketamine more difficult and yield less
  4. Separate the water from the DCM (DCM is water immiscible which is what we are leveraging with this "wash"). You can use a sep funnel, or I find using a micropipette and puling 10ml at a pull easy and effective enough for myself.
  5. Repeat 3 & 4, at least 1 more time. You may repeat again if the water seems to be pulling more molecules from the DCM - i.e., the water is still becoming cloudy relative to initial wash. Note the obvious that the each subsequent wash pulls significantly less stuff from the DCM, as those water agreeable compounds are less present in the DCM.
  6. Dry the wash water in drying tray - use gentle head and vacuum if you have it. A residue forms that seems to contain a good deal of sugar and some waxy/fat-like stuff and, of course the ketamine.
  7. Once completely dry, add 50ml of the freezing anhydrous acetone to the tray containing the dry wash residue.
  8. use brush to lightly scrub the residue... you'll see that the ketamine is there, it's the greater part of the remaining solids. be gentle, use gloved fingers to rub the residue to speed up process.
  9. Use a micropipette to gently and carefully siphon off the acetone and leave the solids, careful of the fine dust-like ketamine - discard the acetone safely
  10. Allow to dry completely free of solvent the acetone washed ketamine residue
  11. Scrape dry residue. IT should be a powder that is mainly and obviously crystalline, a little junk, but pure enough to easily scrape and crush into a powder that looks like powdered sugar.
  12. TA DA. Your rx is undone, and your sugar k is ready, Mr. Wonkman
  13. Do whatever the fuck you want now... I don't condone even reading this

UPDATE: this procedure results in a very crystalline Adderall extraction. And I have not completed with the aqueous alcohol addition to the wash.. amphetamine are more soluble in alcohol than water, so help them get into the water... add isopropyl alcohol - add ISOPROPYL ALCOHOL at 1/10th ratio of the water to make the aqueous wash (If you have 100ml water, add 10ml IPA).
