What happens if an app package is downloaded manually but the same app is made available in Self Service via the Mac App Store at a later time?

In this specific case, I have users who already downloaded an application that I am planning to make available via Self Service, but I want to confirm there won’t be any conflicts.
If there is an update for the Mac App Store version, will it try to update the manually installed package? And if so, would it succeed?
