How do you make an image be opaque based on brightness?

Basically, I have a lineart with a white background and lines that aren't nearly all perfectly black. How can i make the image opaque based on brightness? (I use Sai to color; there's a way to do it in Gimp, but i get errors exporting it. I also have RealWorldCursorEditor and Krita)
