Need Some Help! - NPC "Leveling"

So to give you some background, I made an NPC Transmuter Wizard and Zealot Barbarian. I did this to test out a PC concept of "Dr Jekyll and Mr Hyde", using "wizard" for the doctor and "barbarian" for Hyde. His name is Reginal Wintersong fyi. His goal in life is to "cure his beast like tenancies" and to "discover a way to live forever (aka fountain of youth, elixir of immortality type of thing, thus his "Zealot" Barbarian traits.). He is Currently 3 Wiz/ 3 Barb

So this character visited the City of the Dead during the expanded mission provided in the Megathread for the "Darkling Tattoo Ink" highlighted as side mission type of work. I created my own encounter, making the City of the Dead a place for a hidden casino (gives a "reason" for the Darkling to be there). Anyway, moving into chapter 3 fairly soon and I'm wanting to create some plot hooks to help him "discover" some of these things. Being a Transmuter, at level 6 he'd get a Transmuter Stone which feels like its on the verge of becoming Philosopher's Stone like. The trouble is finding a "hook" and a way for the Wizard to "learn" about these new ways in a fun and interesting way so he can get to the level of "Transmuter Stone" and have a "base" understanding of how it works and work his way up to "immortality". I'm a new DM and I'm trying to go out of my element to try new things and experiment with stuff I have never gotten to as a PC and I'd like to have something just in case Reginald approaches the group or if the group approaches him and is like "So how goes the work" ect ect ect.
