I've never heard that advice from the reagent vendors or any other knowledgeable people. I assume that, because the reagents are so sensitive, they can pick up very small amounts of reactive compounds. It seems to me like it would be difficult for a pill producer to avoid small amounts of the drug in every corner of the pill even if they tried.
Though I suppose there'd be no harm in crushing the pill and mixing it well before pulling a sample to test.
