Seems like I have forgotten to perform an experiment.

Has anyone ever faced a problem like they are unable to perform an experiment which never failed before? Like you could perform the experiment before having no problem in getting positive results but suddenly nothing is working? You have changed all the chemicals and reagents but cannot seem to make the reaction work? I had no problem in extracting DNA from insects a year ago but now I cannot seem to get it right no matter what I try. Its getting frustrating. I have been stuck for a month.
