From the Rolling Stones interview we now know the year the speech was made by JFK, it was 1961, and we know it is unedited.
So what speech could it be?
I'm betting on his inauguration speech ("ask not what your country can do for you , but for what you do could do for your country"). Issue is that the speech is 16 minutes long. I read unedited as not a excerpt but the whole quote.
Other contenders are the famous "moon shot" speech, or a speech given to the press publications.
I'm mobile at the moment, so maybe someone could link transcripts of these speeches so we could maybe figure it out?
