What are the best challenge runs to do in Fallout 3 when you know the game inside and out?

I am looking for a fun challenge run, preferably roleplay based, to do in Fallout 3. I know every nook and cranny in the Capital Wasteland and know almost every line of every character in it. Yes, like most of you, I've played Fallout 3 for thousands of hours.
But, it is still my favourite Fallout so far and the only one made by Bethesda. So I want to play it again before Fallout 4 comes out. This is why I'm looking for a good challenge run idea.
