Being overly technical like you would expect a grade school teacher to be.
"Can I come in" - is asking: "am I physically capable of entering"
"May I come in" - is asking "Do I have your permission to enter."
But it is very uncommon for someone to be this pedantic. Even in formal settings. (At least where I'm from.)
For the joke, a vampire can't enter without permission of the owner, but instead of asking permission the vampire is asking if they have the ability to enter. To which the teacher is mockingly asks the vampire if he can enter.
