Setting Gain With A Multimeter

I have a Kenwood KFC W12PS 12” (250 Watts RMS) bridged with a Kenwood KAC 6202 amp (200 Watts RMS Bridged) and was wondering if the process for setting my gain with a multimeter is the same even though my sub RMS is slightly higher than my amp. I got 28.28 Volts as my optimal voltage so do I just turn my gain up with the volume set to the highest distortion free level on my head unit until my multimeter reads 28? Am I doing this right?

Also can I use the first level of my head units bass boost before tuning and it still be safe as long as I don’t exceed the Voltage?
