Masters first?

I am gathering up information about grad schools and am struggling to decide if I should go for a masters in biochemistry first or just jump straight to PhD. I will go for a PhD eventually, and I am aware that one usually obtains a masters during said PhD. However, as it stands, I am just unsure of my overall ability to do so right now.

I have been working at a good university in a highly regarded PI's lab for going on 3 years and can obtain numerous letters of recommendation from my colleagues. My issue is that I was unprepared for this line of work after undergrad, as I obtained a degree that did not match with what my aspirations became by the end (it was environmental science). I was just trying to graduate at that point. After a while, I eventually landed my current position in a field that I had zero prior experience in (mass spectrometry if that detail helps). It has been a long while since then and my skillset/overall aptitude has greatly increased, however I am still unsure about my ability to conduct a study from beginning to end by myself that a PhD would require. It seems like a large leap, and I am wondering if getting a masters in biochemistry first would be a good choice to shore up some the skillsets I'd need?
