How do you identify and isolate a gene or enzyme?

I'm currently reading about the fungi that can enzymatically decompose the components in crude oil. This was identified by growing the fungus near a puddle of crude oil and watching it grow towards it. My question is, from there, how do you identify the gene or enzyme responsible? I know how to purify proteins and characterise them, but not identify them from scratch.
