X grams / kg calculations seem overdone

It seems different to me that with certain chemicals such as alcohol, that body size has a significant role in effect, but with others such as acid or MDMA, it is more a brain chemistry issue than body size. I see very large men trip way harder than small women on the same dose from the same source, at the same time, over and over (meaning it's not a set and setting issue).
Do you agree?
If so, why don't we hear more about this?
